چکیده

BVDV, currently classified in the pestivirus genus of the family Flaviviridae, are small, enveloped viruses having a single-stranded positive-sense RNA genome. The BVDV genome is approximately 12.3 kb in length with a 5'-nontranslated region (NTR), a single large open reading frame (ORF), and a 3'-NTR lacking a poly(A)tail. The 5'-NTR contains an internal ribosome entry site that initiates translation of BVDV mRNA in a cap-independent manner. The secondary structure of the 5'-NTR is involved in the regulation of both translation and genome replication. The ORF is translated into a single polyprotein of approximately 4000 amino acids that is co-and post-translationally cleaved into 11 or 12 mature proteins by viral and host proteases. The order of proteins in the polyprotein is Npro (a nonstructural autoprotease unique to pestiviruses), the capsid protein (C), the envelope glycoproteins (Erns, E1, and E2), and the nonstructural proteins (p7, NS2/NS3, NS4A, NS4B, NS5A, and NS5B). NS3 (NS2/NS3) has helicase, serine protease, and NTPase activities, and NS5A has RNA-dependent RNA-polymerase activity. The NS2/NS3 complex is not cleaved in BVDV isolates that are noncytopathic in cell culture, while in isolates that are cytopathic both NS2/NS3 and a discrete NS3 are observed. The processing of NS2/NS3 appears to develop from RNA recombination events during the genomic replication of a noncytopathic virus. As a result, the genome of cytopathic isolates may contain genomic duplications, deletions, rearrangements, and/or insertions of cellular mRNA.

Ribosomal S27a coding sequences upstream of ubiquitin coding sequences in the genome of a pestivirus.

Molecular characterization of cytopathogenic (cp) bovine viral diarrhea virus (BVDV) strain CP Rit, a temperature-sensitive strain widely used for vaccination, revealed that the viral genomic RNA is about 15.2 kb long, which is about 2.9 kb longer than the one of noncytopathogenic (noncp) BVDV strains. Molecular cloning and nucleotide sequencing of parts of the genome resulted in the identifica...

Complete Genome Sequence of a Bovine Viral Diarrhea Virus Strain Isolated in Southern China

We report here the full-length RNA genomic sequence of the bovine viral diarrhea virus (BVDV) strain GX4, isolated from a cow in southern China. Studies indicate that BVDV GX4 belongs to the BVDV-1b subtype. This report will help in understanding the epidemiology and molecular characteristics of BVDV in southern China cattle.

BVDV induced gastro-neuropathy outbreak in a feedlot calves around Tehran (Iran)

An outbreak of a lethal disease was reported in 4–6-month-old Holstein calves in a feedlot around Tehran. The signs of central nervous system and gastrointestinal system (GI) involvement were observed in the diseased animals. Necropsy samples of GI, liver, kidney, spleen and lung from 3 died animals were prepared for histopathological examination. Blood and formalin-fixed ear notch samples of 6...

Genoepidemiological evaluation of Bovine viral diarrhea virus 2 species based on secondary structures in the 5' untranslated region.

Bovine viral diarrhea virus 2 (BVDV-2) strains demonstrated in cattle, sheep, and adventitious contaminants of biological products have been evaluated by the palindromic nucleotide substitutions (PNS) method at the three variable loci (V1, V2 and V3) in the 5' untranslated region (UTR), to determine their taxonomical status. Variation in conserved genomic sequences was used as parameter for epi...

Authentic and chimeric full-length genomic cDNA clones of bovine viral diarrhea virus that yield infectious transcripts.

Bovine viral diarrhea virus (BVDV) is the most insidious and devastating viral pathogen of cattle in the United States. Disease control approaches must be based on detailed knowledge of virus biology. To develop reverse-genetic systems to study the molecular biology of the virus, we first constructed a plasmid containing the entire genome of BVDV cloned as cDNA. Subsequently, we showed that inf...
